Background

FGFR2 is a cell-surface tyrosine-kinase receptor for fibroblast growth factors that governs proliferation, differentiation, migration, and apoptosis during embryonic patterning, limb and lung morphogenesis, osteogenesis, and skin development. In osteoblasts, it differentially promotes proliferation/apoptosis depending on maturation stage. Ligand-activated FGFR2 phosphorylates PLCG1, FRS2, and PAK4, triggering PLCγ–DAG/IP3 and GRB2/GAB1/PI3K/SOS1–RAS/MAPK/AKT cascades. Signaling is attenuated by ubiquitination-mediated internalization and degradation; mutations causing constitutive activation or impaired trafficking yield aberrant signaling, while overexpression enhances STAT1 activation.
