Background

Thymic stromal lymphopoietin (TSLP) is an interleukin-7 (IL-7)-like cytokine primarily expressed by epithelial cells such as keratinocytes. The human TSLP cDNA encodes a 159-amino acid precursor protein containing a 28-amino acid signal peptide. TSLP exhibits dual core functions: on one hand, it plays a key role in initiating type 2 immune responses by acting on its most well-characterized target cells—CD11c+ dendritic cells. It promotes TH2 cell responses and directly enhances the effector functions of CD4+ TH2 cells and basophils, thereby driving the allergic inflammation associated with diseases such as atopic dermatitis and asthma. On the other hand, TSLP also contributes to maintaining immune balance, including establishing non-deletional central tolerance, maintaining a non-inflammatory intestinal environment, and limiting pro-inflammatory cytokines. Consequently, TSLP serves as a pivotal molecular link between the epithelial barrier and the immune system, involved both in host defense against helminth parasites and in the pathogenesis of various inflammatory diseases.
