Background

PIK3CD is a gene that encodes a protein which is an integral part of the phosphatidylinositol 3-kinase (PI3K) signaling pathway. This pathway is crucial for various cellular functions including cell growth, proliferation, survival, and metabolism. The PIK3CD protein specifically refers to the delta isoform of the catalytic subunit of PI3K. It is predominantly expressed in immune cells and plays a significant role in the regulation of immune responses. Abnormal activation or mutations in the PIK3CD gene can lead to a range of diseases including certain types of leukemia and other immune-related disorders. Understanding its function and regulation is important for developing targeted therapies in cancer and immune system disorders.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human colon. Anti-PIK3CD antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

IHC shows positive staining in paraffin-embedded human tonsil.

IHC shows positive staining in paraffin-embedded human colon cancer.

IHC shows positive staining in paraffin-embedded human gastric cancer.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in Jurkat cells. Anti- PIK3CD antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
