Background

Mouse interleukin-4 (IL-4) is a pleiotropic cytokine that plays a central role in regulating immune responses, primarily produced by activated T helper 2 (Th2) cells, mast cells, and basophils. It is a key driver for the differentiation of naive CD4+ T cells into Th2 cells and promotes the proliferation and differentiation of B cells. The protein also regulates immunoglobulin class switching, enhancing the production of IgG1 and IgE, and induces the expression of MHC class II molecules. Mouse IL-4 exerts its effects by binding to a heterodimeric receptor complex composed of the IL-4Rα chain and the common gamma chain (γc), which activates the JAK/STAT6 signaling pathway. Recombinant mouse IL-4 is typically a non-glycosylated polypeptide chain consisting of approximately 120-121 amino acids, with a molecular weight around 13.5-14 kDa. It is commonly produced in E. coli expression systems for research purposes, where its biological activity is often confirmed by promoting the proliferation of HT-2 cells. Given its critical functions in Th2-mediated immunity, allergic responses, and parasitic infections, mouse IL-4 is an essential tool for studying these pathways in preclinical models.
