Background

COXIV is a key subunit of cytochrome c oxidase (Complex IV), the final enzyme complex in the mitochondrial respiratory chain. This protein consists of approximately 160 amino acids, containing an N-terminal mitochondrial targeting sequence and multiple α-helical transmembrane domains, and is primarily anchored in the inner mitochondrial membrane. COXIV serves dual important functions—structural and regulatory: on one hand, it provides structural support for the core catalytic subunits (COX I and COX II) of Complex IV, assisting in the assembly and stability of the entire enzyme complex; on the other hand, it senses the cellular energy state through an ATP/ADP binding site, thereby allosterically regulating the catalytic activity of Complex IV to match the efficiency of electron transfer in the respiratory chain with ATP synthesis demands. Furthermore, COXIV expression levels vary across different tissues (such as the liver-type COXIV-1 and lung-type COXIV-2 isoforms), and its expression is regulated by transcription factors such as hypoxia-inducible factor HIF-1α, playing a role in cellular responses to hypoxic stress and metabolic reprogramming. At the pathological level, mutations or aberrant expression of COXIV are closely associated with various mitochondrial diseases, including mitochondrial encephalomyopathies and Leigh syndrome. Its dysfunction leads to decreased electron transport chain efficiency and excessive production of reactive oxygen species (ROS), making COXIV also an important potential target in research on ischemia-reperfusion injury, neurodegenerative diseases, and tumor metabolism.
