This all-in-one 4-stage kit enables 3D cardiac organoid differentiation, fully recapitulating the in vivo developmental microenvironment of the human heart. No separate preparation of growth factors or small-molecule inhibitors is required. The standardized 20-day differentiation workflow generates self-cavitated, spontaneously contracting human cardiac organoids.
Differentiated products contain multiple native cardiac cell populations: atrial/ventricular cardiomyocytes, epicardial cells, endothelial cells and cardiac fibroblasts. The cell composition and spatial architecture closely mimic early human cardiac tissue. The organoids support long-term maturation culture, electrophysiological assays, cardiotoxicity testing of pharmaceuticals, in vitro modeling of congenital heart diseases, and mechanistic research on cardiac development.

Day 0–Day 2: Stage I medium induces mesoderm formation; U-bottom ultra-low-adhesion 96-well plates support spontaneous 3D embryoid body generation.
Day 3–Day 4: Stage II medium directs cardiac mesoderm specification.
Day 5–Day 6: Stage III medium drives cardiomyocyte lineage differentiation.
Replace with Stage IV maturation medium starting on Day 7. Mature contracting cardiac organoids are obtained by Day 20. Extended culture up to 35+ days is available for functional assays.
