IF shows positive staining in paraffin-embedded human colon cancer. Anti- CEA (Alexa Fluor® 700 Conjugate) antibody was used at 1/100 dilution (magenta) and incubated overnight at 4°C. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.

Background
Carcinoembryonic antigen (CEA) is a glycoprotein produced in colorectal cancer tissue. In normal adult tissues, CEA is expressed at the apical border and weakly expressed in the cytoplasm, such as columnar cells of the colon, small intestine, stomach (surface epithelium, cervical mucus cells and weak expression in pyloric mucus cells), pancreatic ducts, secretory epithelium of sweat glands, squamous epithelium of tongue, esophagus, cervix and urothelium. CEA antibodies aid in the classification of adenocarcinomas, particularly in the gastrointestinal tract, including colon and pancreatic cancers. IHC results can also help differentiate between secretory meningiomas and medullary thyroid carcinomas.
