Background

Ag85B is a major secreted antigen and mycolyltransferase of the Mycobacterium tuberculosis complex. This multifunctional enzyme plays a central role in constructing the unique mycobacterial cell wall. As part of the Ag85 complex, it catalyzes the transfer of mycolic acids—long-chain fatty acids—onto the arabinogalactan-peptidoglycan matrix, forming the essential mycolyl-arabinogalactan-peptidoglycan complex (mAGP). This creates the robust, hydrophobic outer membrane critical for pathogenicity and antibiotic resistance. Due to its high immunogenicity, Ag85B is also a leading candidate antigen in several tuberculosis vaccine strategies.
