Background
Rat Interleukin-6 (IL-6) is a pleiotropic cytokine encoded by the Il6 gene on chromosome 4 (NC_086022.1, 5889999..5894575, complement), consisting of 5 exons that produce a protein-coding transcript . As a key member of the IL-6 family, it orchestrates acute-phase responses, immune regulation, and metabolic homeostasis in rodents. Signals through two distinct pathways in rats: Classic signaling: IL-6 binds membrane-bound IL-6Rα, recruiting gp130 to activate JAK1/STAT3 cascade; Trans-signaling: Soluble IL-6Rα (sIL-6R) complexes with IL-6 to activate gp130 on cells lacking membrane IL-6Rα, amplifying inflammatory responses in neurons and vascular tissue.
