Background

Insulin-like Growth Factor Binding Protein-1 (IGFBP-1) is a 25–30 kDa secreted protein encoded by the Igfbp1 gene, predominantly expressed in the liver, decidua, and kidneys. As a critical regulator of the IGF axis, IGFBP-1 binds IGF-I and IGF-II with high affinity (Kd ~10⁻¹⁰ M), thereby modulating ligand bioavailability, half-life, and receptor interactions.

IGFBP-1 in mouse models represents a critical nexus between growth regulation and metabolic homeostasis, with applications spanning developmental biology, metabolic disease modeling, cancer resistance mechanisms, and cardiovascular protection. Industrial applications focus on biomarker assay development, RGD-based peptide therapeutics, and combination strategies to overcome cancer drug resistance.

Bioactivity

Measured by its ability to inhibit the biological activity of IGFII on MCF7 human breast adenocarcinoma cells. The EC50 for this effect is 15-25 ng/mL in the presence of 14 ng/mL human IGFII.
