Background

TOP2B (DNA topoisomerase II beta) is a key nuclear enzyme ubiquitously expressed in eukaryotic cells, belonging to the type II topoisomerase family. It catalyzes the interconversion of DNA topological states by transiently cleaving and religating DNA double strands, thereby relieving supercoiling and catenation generated during DNA replication, transcription, and recombination. Unlike TOP2A, which primarily functions in DNA replication during cell proliferation, TOP2B is expressed in both non-proliferating and proliferating cells, and its activity is closely associated with gene transcription regulation—particularly during differentiation processes such as neurodevelopment, hematopoiesis, and adipogenesis, where it maintains tissue-specific gene expression programs by modulating chromatin loop structures and long-range enhancer-promoter interactions. Additionally, TOP2B participates in DNA damage repair and chromatin remodeling, and its dysfunction can lead to genomic instability. At the clinical level, TOP2B is an important target for various anticancer drugs (such as etoposide and doxorubicin), which induce DNA breaks by stabilizing the DNA-TOP2B cleavable complex, thereby killing tumor cells. However, this mechanism may also cause side effects such as therapy-related leukemia, and TOP2B dysregulation has also been associated with neurodegenerative diseases and certain developmental disorders.
