Background

LIF (Leukemia Inhibitory Factor) is a multifunctional pleiotropic cytokine belonging to the IL-6 cytokine family. It binds to a composite receptor on the cell surface composed of LIF receptor (LIFR) and gp130 subunits, activating multiple downstream signaling pathways including JAK/STAT3, PI3K/AKT, and MAPK, thereby exerting a wide range of biological functions across various tissues. It is critically important in embryonic development, serving as the classic "stem cell maintenance factor" that maintains mouse embryonic stem cell pluripotency and prevents their differentiation. In the nervous system, it promotes the survival of neural progenitor cells and the differentiation of astrocytes, while also playing a role in promoting repair and regeneration after nerve injury. Furthermore, LIF is deeply involved in immune regulation (such as promoting Th17 cell differentiation), bone metabolism regulation, inflammatory responses, and reproductive physiological processes like endometrial decidualization. Therefore, aberrant LIF expression is closely associated with various diseases, including certain autoimmune disorders, osteoarthritis, neurological diseases, and tumor development and progression. Clinically, recombinant LIF has also been explored for improving embryo implantation to enhance assisted reproductive outcomes.
