Background

HLX protein (H2.0-like homeobox protein) is an evolutionarily highly conserved transcription factor belonging to the H2.0 homeobox family. It regulates the expression of downstream genes through its sequence-specific DNA-binding ability and functions on chromatin within the nucleus. This protein plays multiple critical roles in the body: during development, it participates in embryonic digestive tract morphogenesis, enteric nervous system development, and the growth of vital organs such as the liver; in the immune system, it serves as a key factor in promoting Th1 cell differentiation and maintaining their function, which is essential for coordinating immune responses; additionally, in endothelial cells, this protein is regulated by the vascular endothelial growth factor (VEGF) signaling pathway and upregulates the expression of guidance molecules such as UNC5B, thereby inhibiting excessive endothelial sprouting and migration, acting as a "brake" in the fine-tuning of angiogenesis. Consequently, HLX dysfunction is associated with various pathological processes, involving acute myeloid leukemia, placental developmental abnormalities, and congenital diaphragmatic hernia, among other diseases.
