Background
TSLP (thymic stromal lymphopoietin) is a four-helix-bundle cytokine of the IL-2 family that is chiefly produced by epithelial cells at barrier surfaces; it acts on dendritic cells, mast cells, and type 2 innate lymphoid cells via a receptor complex of TSLPR plus IL-7Rα to trigger STAT5-driven transcriptional programs, thereby polarizing naïve CD4⁺ T cells toward a pro-allergic Th2 phenotype, orchestrating eosinophilic inflammation, and serving as a master switch for atopic dermatitis, asthma, and other allergic diseases.
