Flow cytometric analysis of Mouse CD196 expression on C57BL/6 mouse splenocytes. C57BL/6 mouse splenocytes were stained with Pacific Blue™ Rat Anti-Mouse CD45R/B220 Antibody and either Biotin Armenian hamster IgG Isotype Control (Left panel) or SDT Biotin Armenian hamster Anti-Mouse CD196 Antibody (Right panel) at 5μl/test followed by Sav-PE. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD196, also known as CCR6, is a ~42 kDa G protein-coupled seven-transmembrane chemokine receptor expressed on immature dendritic cells, memory T and B cells, NK cells and Langerhans cells; it binds the unique ligand CCL20 (MIP-3α), triggering calcium signaling that directs chemotactic recruitment of these cells to epithelial and inflamed tissues, thereby orchestrating mucosal immunity, Th17 and regulatory T-cell differentiation, B-lineage maturation, and pathogenic inflammatory circuits in diseases such as psoriasis, rheumatoid arthritis and inflammatory bowel disease.
