Background

Trypsin is a serine protease commonly found in the vertebrate digestive system that hydrolyzes proteins and is secreted in the pancreas as inactive trypsinogen. Its site of cleavage of the peptide chain is located primarily at the carboxyl terminus of lysine or arginine (except when both are immediately followed by proline). Trypsin consists of two subunits, the alpha subunit (with 2 polypeptide chains forming it) and the beta subunit (with 1 polypeptide chain forming it). Its hydrolytic activity can be inhibited by a variety of inhibitors, including 1) organophosphorus compounds, such as isopropyl fluorophosphate; 2) natural trypsin inhibitors derived from pancreas, soybeans, green beans, and egg whites; 3) silver ions; and 4) specific protease inhibitors, such as AEBSF, anti-protease, peptidylase, DFP, leucoinhibitory peptidase, PMSF, TLCK, etc.

Components

Trypsin,Mass Spectrometry Grade lyophilized
2*Reaction buffer:,100 mM Tris-HCl 、40 mM CaCl2 pH 8 @ 25°C

Protocol

1 Trypsin, Mass Spectrometry Grade should be reconstituted by the addition of 20–200 μl of 2mM HCl
2 Set-up a typical reaction as follows
1)Add the following components in sequence

Components

Volume

Substrate protein

5 μl (about 2 ug)

ddH2O

4.5μl

2*Reaction buffer

10μl

100 ng/μl Tyrpsin

0.5 μL

2)Incubate at 37°C for 2~4h

Guidelines

1. Substrate must be in phosphate-free buffer to prevent calcium precipitation with both reconstituted enzyme and enzyme buffer.

2. Storage buffer:2mM HCl, pH3.0.

Unit Definition

In a 3.2ml reaction system using BAEE as substrate, the UV absorption at 253nm increased by 0.003 per min using cuvette with 1 cm optical path, which was defined as 1 USP trypsin activity unit

Picture

Bioactivity
