Background
TPH-1 (tryptophan hydroxylase-1) is a 51-kDa, biopterin-dependent and rate-limiting enzyme that catalyzes the hydroxylation of L-tryptophan to 5-hydroxytryptophan, thereby initiating peripheral serotonin synthesis in enterochromaffin cells, platelets, and mast cells; it is a homodimer whose activity is regulated by phosphorylation, allosteric cAMP-dependent mechanisms, and interaction with 14-3-3 proteins, and genetic or post-translational dysregulation of TPH-1 has been implicated in gastrointestinal disorders, cardiovascular fibrosis, and mood-related diseases.
