Background

Sterol regulatory element-binding protein 2 (SREBP2) is a membrane-bound transcription factor that acts as the master sensor and amplifier of cellular cholesterol demand: in cholesterol-replete ER membranes it is held inactive through association with SCAP and Insig, but when cholesterol falls SCAP escorts SREBP2 to the Golgi where site-1 and site-2 proteases release its ~480-amino-acid N-terminal fragment; this mature form enters the nucleus, binds sterol response elements, and drives expression of nearly every gene required for cholesterol acquisition (HMGCR, HMGCS, LDLR) and efflux control (ABCA1, ABCG5/8), while also cross-talking with SREBP1c to coordinate overall lipid homeostasis, making SREBP2 the central switch that keeps mammalian cell membranes optimally functional.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-SREBP2 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
