Background

RBPMS (RNA-binding protein with multiple splicing) is an RNA-recognition-motif protein that binds tandem CAC motifs in pre-mRNAs to orchestrate tissue-specific alternative splicing, mRNA stability and translational control; in the heart it regulates inclusion of sarcomeric exons (e.g., Ttn exon 47, Pdlim5, Nexn) essential for post-natal contractility, so its loss causes dilated cardiomyopathy and heart failure, while in retina and neurons it serves as a selective marker of retinal ganglion cells and modulates axonal integrity, and during development it governs cardiomyocyte cytokinesis, preventing premature binucleation and non-compaction cardiomyopathy, thereby exerting stage- and cell-type-specific control over RNA processing critical for vision, neural function and cardiac performance.

Immunohistochemistry

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

IF shows positive staining in paraffin-embedded mouse retina. Anti-RBPMS antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue).
