Background
RANKL (Receptor Activator of Nuclear Factor Kappa-B Ligand), also known as TNFSF11, is a critical type II transmembrane protein belonging to the TNF superfamily that functions as the principal stimulatory factor for osteoclast formation, activation, and survival by binding to its receptor RANK on osteoclast precursor cells, thereby playing an essential role in bone remodeling, calcium metabolism, and immune responses, while its overexpression is implicated in various bone-related diseases such as osteoporosis, rheumatoid arthritis, and bone metastases, making it a key therapeutic target for denosumab and other RANKL inhibitors.
