Background
Phospho-ULK1 (Ser757) is the phosphorylated form of the serine/threonine kinase ULK1 at serine 757, a modification catalyzed by mTORC1 under nutrient-rich conditions that disrupts ULK1’s interaction with AMPK and keeps autophagy suppressed; upon starvation or rapamycin-triggered mTORC1 inhibition, Ser757 dephosphorylation enables AMPK binding and subsequent phosphorylation of activation-loop sites, licensing ULK1 kinase activity and initiating autophagosome formation.
