Background

Phospho-Glycogen Synthase (Ser641) is the inactive, phosphorylated form of glycogen synthase in which a phosphate group is covalently attached to serine-641; this single modification, catalyzed chiefly by glycogen synthase kinase-3 (GSK-3) downstream of insulin signaling, sterically hinders the enzyme’s active site and prevents the elongation of glycogen chains, thereby providing a rapid, reversible switch that links hormonal cues to systemic glucose storage.
