Background

Phospho-Bad (Ser112) is the phosphorylated form of the pro-apoptotic Bcl-2 family member Bad at serine 112, a modification catalyzed by survival kinases such as PKB/Akt, p90RSK or Pim-2 that creates a 14-3-3-binding motif, sequestering Bad in the cytosol and thereby preventing it from neutralizing anti-apoptotic Bcl-2/Bcl-xL at the mitochondrial outer membrane; this single phosphorylation event thus functions as a molecular switch converting Bad from an apoptosis inducer to a silent scaffold, integrating growth-factor, stress and cytokine signals to set the apoptotic threshold in neurons, lymphocytes and many cancer cells.
