Background

MAPK13 and MAPK14, commonly known as p38δ and p38α respectively, are critical members of the p38 mitogen-activated protein kinase (MAPK) signaling pathway. Functioning as essential intracellular signaling molecules, they are primarily activated in response to diverse stress stimuli, such as inflammatory cytokines, environmental stress, and oxidative stress. Their activation depends on the dual phosphorylation of specific threonine and tyrosine residues, which in turn regulates the activity of numerous downstream transcription factors and proteins. MAPK14 (p38α) is the most extensively studied and ubiquitously expressed isoform, playing a central role in processes like inflammatory responses, apoptosis, cell differentiation, and cell cycle regulation, making it a significant drug target for inflammatory and autoimmune diseases. In contrast, MAPK13 (p38δ) exhibits a more tissue-specific expression pattern, predominantly found in glandular tissues (e.g., salivary glands, pancreas), skin, and lungs, where it fulfills distinct roles in tissue-specific inflammation, skin pathophysiology, and the regulation of insulin secretion. Although they share upstream activators and some downstream substrates, MAPK13 and MAPK14 exhibit both overlapping and complementary physiological functions, collectively forming a core network for the precise cellular regulation of stress responses.

Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
