Background

Milk fat globule-EGF factor 8 (MFG-E8), also called lactadherin, is a secreted 66–75 kDa glycoprotein composed of an N-terminal EGF-like domain containing an RGD integrin-binding motif and two C-terminal discoidin/F5-8 domains that bind phosphatidylserine, enabling it to act as a bridging molecule between apoptotic cells and phagocytes expressing αvβ3/αvβ5 integrins, thereby orchestrating efferocytosis, dampening inflammation, and exerting pleiotropic effects on vascular remodeling, skeletal-muscle lipid metabolism, myogenic differentiation, osteoclastogenesis, and tumor progression while also giving rise to the amyloidogenic peptide medin found in aged arterial walls.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human liver. Anti- MFG1-E8 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in PANC-1 cells. Anti- MFG1-E8 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
