Background
LILRB4 (Leukocyte Immunoglobulin-Like Receptor B4), also known as ILT3, is a type I transmembrane protein with two extracellular Ig-like domains and three intracellular ITIM motifs that recruit SHP-1/2 phosphatases to dampen myeloid-cell activation; expressed chiefly on monocytes, macrophages and dendritic cells, it functions as a key immune checkpoint that maintains self-tolerance by inhibiting NF-κB/MAPK signaling, suppressing T-cell proliferation and promoting Treg/Ts differentiation, but is exploited by tumors such as AML through ligands like ApoE to enable immune evasion, leukemia invasion and poor prognosis, making it an emerging therapeutic target in cancer, autoimmunity and transplantation.

IHC shows positive staining in paraffin-embedded human tonsil. Anti-LILRB4 antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
