Background
HPV16 E7 is a 98-amino acid key oncoprotein of human papillomavirus (HPV) type 16; via its conserved CR1/CR2 motifs it binds the retinoblastoma tumor suppressor pRb and related pocket proteins, displacing E2F transcription factors and forcing the infected epithelial cell into S-phase, leading to cell cycle deregulation and unregulated abnormal proliferation. And E7 protein can further enhance the malignant phenotype of cells by binding to other cell cycle inhibitory proteins such as p21 and p27, or activating signaling pathways that promote cell survival and migration, such as PI3K/Akt and MAPK. It also inhibits the normal death of damaged cells, accumulates mutations, and drives the occurrence and progression of HPV-related cancers.
