Background
Glycogen Synthase (GS) is the key rate-limiting enzyme in the glycogen biosynthesis pathway, primarily responsible for catalyzing the elongation of the glycogen chain. It uses uridine diphosphate glucose (UDP-Glucose) as the activated glucose donor, adding glucose molecules via α-1,4-glycosidic bonds to a pre-existing glycogen primer chain, thereby linearly extending the glucose polymer. The activity of this enzyme is precisely regulated by both allosteric control and covalent modification (reversible phosphorylation): the insulin signaling pathway activates its activity through dephosphorylation, promoting glycogen storage; whereas stress hormones like glucagon and adrenaline cause its inactivation through phosphorylation, thereby inhibiting glycogen synthesis and mobilizing energy release. Thus, Glycogen Synthase is a central molecule in maintaining the body's glucose homeostasis, and its aberrant activity is closely associated with metabolic diseases such as Type II diabetes and insulin resistance.

IHC shows positive staining in paraffin-embedded human colon. Anti-Glycogen Synthase antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
