Background

ERAP1 (Endoplasmic Reticulum Aminopeptidase 1) is a zinc-dependent metalloaminopeptidase localized in the endoplasmic reticulum that plays a critical role in antigen processing by trimming N-terminal residues of peptides to optimize their length (8–10 amino acids) for loading onto MHC class I molecules, thereby shaping the immunopeptidome and influencing immune responses; it also modulates inflammation by cleaving cytokine receptors and is genetically associated with autoimmune diseases such as ankylosing spondylitis and Behçet’s disease, particularly in individuals carrying specific HLA alleles like HLA-B27 or HLA-B*51.
