Background
Cyclin E1 is a regulatory subunit of the cyclin-dependent kinase Cdk2 that drives the G1-S phase transition by phosphorylating substrates such as the retinoblastoma protein, thereby releasing E2F transcription factors and committing cells to DNA replication; its expression peaks in late G1, is tightly controlled by ubiquitin-mediated proteolysis, and is frequently amplified or overexpressed in cancers, leading to genomic instability and aggressive tumor behavior.
