Background

CD69 is a type II transmembrane C-type lectin protein encoded by the CD69 gene, located on chromosome 12 in humans. It is a homodimer composed of two differentially glycosylated subunits (28–32 kDa), each containing an extracellular C-type lectin domain (CTLD), a transmembrane region, and a short cytoplasmic tail. CD69 is an early activation marker rapidly upregulated on the surface of T cells, B cells, and natural killer (NK) cells after stimulation. Its expression is regulated by transcription factors such as NF-κB, AP-1, and ERG-1. Functionally, CD69 plays a crucial role in lymphocyte proliferation, cytokine production, and retention in lymphoid tissues by interacting with molecules like S1P1 and Myl9/12. It is also implicated in the regulation of immune responses in various inflammatory and autoimmune conditions.

Flow cytometric analysis of CD69 expression on unstimulated and stimulated Mouse splenic leukocytes. C57BL/6 Mouse splenocytes were either not stimulated (Left panel) or stimulated overnight (Right panel) with 3 μg/ml Concanavalin A and then stained with either PercP-Cy5.5 Armenian hamster IgG Isotype Control (Black line histogram) and SDT PercP-Cy5.5 Armenian hamster Anti-Mouse CD69 Antibody (Red line histogram) at 5 μl/test followed by Sav-iFluor 488. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
