Background

Perforin is a crucial protein involved in the immune response, primarily produced by cytotoxic T lymphocytes and natural killer cells. It functions as a pore-forming protein, inserting itself into the membranes of target cells, such as virus-infected or cancerous cells. Once integrated, perforin creates pores that disrupt the cell membrane's integrity, leading to osmotic lysis and ultimately killing the target cell. This mechanism is essential for the immune system's ability to eliminate harmful cells and maintain homeostasis.

Flow cytometric analysis of Perforin expression in PBMC (Human peripheral blood mononuclear cells). Human PBMC were fixed and permeabilized with Foxp3 / Transcription Factor Staining Buffer Set. The cells were then stained with Brilliant Violet 421™ Mouse Anti-Human CD8 Antibody and either Alexa Fluor® 488 Mouse IgG2b, κ Isotype Control or SDT Alexa Fluor® 488 Mouse Anti-Human Perforin Antibody at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
