IFN-gamma ELISA Kits: Methodological Design and Research Applications in Type II Interferon Quantification
Why IFN-γ Matters Beyond a Single Cytokine
Interferon-gamma (IFN-γ) is the sole type II interferon and one of the most consequential effector molecules of cell-mediated immunity. It is secreted predominantly by activated CD4+ Th1 cells, CD8+ cytotoxic T cells and NK cells, and it signals through the IFNGR1/IFNGR2 receptor pair to engage the JAK1/JAK2–STAT1 axis. Downstream of that axis, IFN-γ upregulates MHC expression, enhances antigen presentation and potentiates the microbicidal activity of macrophages.
Because it sits at the intersection of infection immunology, tumour biology and autoimmunity, IFN-γ is rarely measured in isolation. Dysregulated IFN-γ expression accompanies tuberculosis infection, autoinflammatory disease, graft-versus-host disease and the response to cancer immunotherapy. Quantifying it reliably in serum, plasma and cell culture supernatant has therefore become a routine requirement in vaccine immunogenicity assessment, mechanistic studies of infection immunity and monitoring of immunotherapeutic response. The recent expansion of latent tuberculosis diagnostics, CAR-T potency testing and IFN-γ release assays has pushed demand for sensitive, standardised IFN-γ quantification even higher.
The Methodological Logic of a Sandwich IFN-γ ELISA
An IFN-γ ELISA kit is built around a simple principle: specific capture followed by signal amplification. A microplate is pre-coated with a tag antibody, and the anti-human IFN-γ capture antibody carries the matching tag. IFN-γ in the sample binds that capture antibody, a detection antibody then completes the sandwich, and TMB substrate generates a colour signal read at 450 nm. Concentration is directly proportional to signal intensity.
Within that shared architecture, performance parameters determine whether results are trustworthy. Taking the Human IFN-γ OneStep HS ELISA Kit as a worked example, the assay uses a one-step double-antibody sandwich format with a detection range of 0.78–50 pg/mL and a sensitivity of 0.316 pg/mL, validated for serum, plasma and cell culture supernatant. Intra-assay precision is 4.5% and inter-assay precision is 6.0%; recovery is 94% in serum and 97% in plasma.
One detail carries disproportionate weight for cross-study comparison: the dose-response curve of this kit runs parallel to the international standard NIBSC 87/586, allowing unit conversion through the relationship NIBSC (87/586) approximate value (IU/mL) = 0.02 × OneStep Human IFN-γ value (pg/mL). That parallel relationship is what makes data comparable across platforms and laboratories.
Notes
- IFN-γ has a short half-life in vivo, fluctuates with stress and is present at low abundance in most samples — sensitivity and specificity requirements are correspondingly high.
- Cross-reactivity with IL-2, IL-4 and TNF-α should be verified during method validation.
Three Detection Scenarios: Total, Antigen-Specific and Release-Assay Analysis
Although a single assay format underlies most IFN-γ ELISAs, the research questions they serve fall into three practical categories.
Total IFN-γ quantification
Measuring bulk serum or plasma IFN-γ is the most established application, reporting the overall activation state of Th1 immunity in infectious disease, autoimmunity and cancer immunotherapy. In haemodialysis patients, plasma IFN-γ correlates positively with monocyte hyper-responsiveness, suggesting that IFN-γ-driven trained immunity sustains the chronic inflammatory state and that targeting IFN-γ may alleviate immune overactivation. In rheumatoid arthritis, single-cell transcriptomic analysis shows IFN-γ response genes significantly upregulated in monocytes and T cells, linking IFN-γ pathway activation to early pathological progression.
Antigen-specific IFN-γ detection
Antigen-specific readouts are the backbone of vaccine cellular immunogenicity assessment and of immune memory evaluation after infection. In tuberculosis diagnosis, IFN-γ release assays such as QuantiFERON quantify IFN-γ released into whole blood after stimulation with mycobacterial antigens to determine latent infection status. Studies have shown that different brands of IFN-γ ELISA kit differ in repeatability, intermediate precision and accuracy when applied to QuantiFERON culture supernatants — a validated kit is therefore a prerequisite for diagnostic consistency. In vaccine development, IFN-γ secreted after antigen re-stimulation is a key indicator of the strength of a vaccine-induced Th1 response.
IFN-γ release assay support
IFN-γ release assays (IGRA) are the mainstream approach to latent tuberculosis diagnosis and depend entirely on precise quantification of IFN-γ in culture supernatant. The same readout underpins CAR-T potency evaluation, where IFN-γ secretion is a core parameter of engineered T-cell functional activity; human IFN-γ detection kits explicitly intended for CAR-T potency assessment are already available on the market, reflecting the place of IFN-γ ELISA in cell therapy quality control.
Research Applications Across Cellular Immunology
Infectious disease and tuberculosis immunodiagnosis
IFN-γ is the central effector molecule controlling intracellular pathogens. In Mycobacterium tuberculosis infection it activates macrophage killing and promotes granuloma formation, both essential to host defence. IFN-γ release assays that measure IFN-γ released after mycobacterial antigen stimulation have become the standard tool for latent tuberculosis diagnosis. Researchers can quantify IFN-γ in QuantiFERON culture supernatant by ELISA and combine it with CD4+/CD8+ T-cell counts to profile host antitubercular immunity.
Tumour immune microenvironment and immunotherapy response
IFN-γ is central to tumour immune surveillance. Secreted by tumour-infiltrating T cells, it upregulates MHC class I expression on tumour cells, enhances antigen presentation and inhibits angiogenesis. Under anti-PD-1/CTLA-4 therapy, activation of the IFN-γ signalling pathway is an important predictive marker of immune checkpoint inhibitor efficacy; bioluminescent reporter mouse models have been used to track the spatiotemporal dynamics of IFN-γ-expressing cells in real time, showing that anti-PD-1/CTLA-4 treatment significantly enhances intratumoural IFN-γ signalling. IFN-γ ELISA on tumour homogenate and peripheral blood, combined with CD8+ T-cell infiltration density, supports assessment of immunotherapy response potential.
Autoimmune disease and chronic inflammation
IFN-γ plays a genuinely bidirectional role in autoimmunity. On one hand it drives Th1 polarisation and the release of inflammatory cytokines; on the other it contributes to immune tolerance through induction of IDO expression and regulation of T-cell function. In haemodialysis patients, IFN-γ-mediated trained immunity correlates with monocyte hyper-responsiveness, suggesting that sustained IFN-γ signalling under chronic inflammation may aggravate immunopathological damage. Measuring IFN-γ together with STAT1 phosphorylation and downstream target genes helps dissect its contribution to disease activity and immune imbalance.
Vaccine immunogenicity and cellular immunity
IFN-γ is the core indicator of vaccine-induced cellular immunity. In viral-vector, mRNA and subunit vaccine development, antigen-specific IFN-γ secretion determines the strength of the Th1 response. Combining ELISpot or ELISA on re-stimulated PBMC supernatant with polyfunctional T-cell analysis (IFN-γ+/TNF-α+/IL-2+) gives a composite picture of vaccine cellular immunogenicity.
Mesenchymal stem cell immunomodulation and cell therapy
IFN-γ priming is a common strategy for enhancing the immunomodulatory capacity of mesenchymal stem cells (MSC). Low-dose IFN-γ (1 ng/mL) is sufficient to induce pronounced changes in gene expression and chromatin accessibility in MSC, activating immune-response genes while suppressing cell-cycle genes — an epigenetic rationale for optimising MSC therapeutic strategies. Monitoring IFN-γ consumption or secretion in MSC culture supernatant by ELISA supports assessment of priming efficiency and immunomodulatory function.
Selecting the Right IFN-γ Assay Format
Because "IFN-γ ELISA kit" describes a family rather than a single product, format selection follows directly from sample type, expected concentration range and the required detection limit. The ANTBIO IFN-γ portfolio maps onto the scenarios above along two axes: detection range and sensitivity.

Closing the Quality-Control Loop
The quality of IFN-γ ELISA data depends on whether sample handling and assay operation form a complete control loop. On the sample side, serum should be centrifuged within 2 hours of collection to avoid haemolysis and repeated freeze-thaw cycles, and the collection timing of cell culture supernatant must be strictly standardised because IFN-γ accumulates continuously in vitro. On the assay side, reagent equilibration, pipetting precision and incubation conditions jointly determine whether the antigen-antibody reaction reaches completion. Embedding both into a single quality-control system is the basic precondition for reproducible, trustworthy quantitative data.
