• Dbl Homology (DH) Domain: This highly conserved region is the key functional domain that enables VAV1 to act as a phosphorylation-dependent GDP/GTP exchange factor (GEF) for Rho GTPases. It can promote the exchange of Rac/Rho GTPases from GDP (inactive state) to GTP (active state), thereby activating Rho GTPases.
• Plekstrin Homology (PH) Domain: Typically, the PH domain is involved in phospholipid binding and cell membrane localization, helping to target VAV1 to the cell membrane for more effective participation in signal transduction.
VAV1 is a multi-domain protein that not only has guanine nucleotide exchange factor (GEF) activity but also acts as a scaffold protein in GEF-independent signaling pathways.

• Activation of Rac/Rho Family GTPases: The primary GEF-dependent function of VAV1 is to activate Rac/Rho family GTPases. When VAV1 is tyrosine-phosphorylated, its GEF activity is activated, catalyzing the replacement of GDP with GTP on Rho GTPases, thereby converting them from an inactive to an active state.
• Cytoskeletal Reorganization: Activated Rac/Rho family GTPases trigger actin reorganization and F-actin polymerization, which in turn affect cell morphology and movement. For example, in T cells, VAV1 promotes actin polymerization by activating Rac1, facilitating the formation of immune synapses between T cells and antigen-presenting cells (APCs).
• Cell Migration and Adhesion: Through the activation of Rho GTPases, VAV1 regulates integrin-mediated cell adhesion activation and participates in chemokine-mediated cell migration. In macrophages, VAV1 activity is essential for CSF-1-mediated chemotaxis and phagocytosis.
• Receptor Clustering and Signal Transduction: The GEF activity of VAV1 contributes to the clustering of T-cell receptors (TCR) and B-cell receptors (BCR), thereby enhancing receptor-mediated signal transduction.
2.1.1 Role of VAV1 in Antigen Receptor Signal Transduction
VAV1 is a key component of the antigen receptor signal transduction complex, associated with both (A) T-cell receptor (TCR)/CD3 and (B) B-cell receptors. In T cells, SLP76 is recruited to the T-cell transmembrane adapter protein (LAT) via its SH2 domain, and the resulting LAT/SLP76 complex serves as a critical scaffold for other proteins (including VAV1) in the proximal TCR signal complex. VAV1 interacts with other proteins through SH2 domains and proline-rich regions/SH3 domains.
In B cells, the VAV1-involved signal complex includes the SLP76-homologous adapter protein SLP65 (also known as B-cell linker protein), Bruton's tyrosine kinase (BTK), growth factor receptor-bound protein 2 (Grb2), and phospholipase γ (PLCγ) 2. The assembly of these protein complexes activates downstream events. The main GEF-dependent function of VAV1 is to activate Rac/Rho family GTPases. In T and B cells, costimulation through CD28 and CD19, respectively, is crucial for optimal phosphorylation of VAV1 and activation of downstream signaling pathways.
• Scaffold Protein Activity: In GEF-independent pathways, VAV1 primarily functions as a scaffold protein. It binds to other proteins through various adapter domains and participates in the formation of antigen receptor-proximal signal complexes. For example, the interaction between VAV1 and phospholipase C-γ (PLCγ) 1 is crucial for TCR-mediated calcium release and activation of the NFAT pathway.
• Promotion of Cell Survival and Cell Cycle Progression: VAV1 promotes cell survival and cell cycle progression in a GEF-independent manner, thereby facilitating the DNA damage response.
• Regulation of Oxidative Stress Response: VAV1 is also involved in regulating the oxidative stress response, influencing intracellular redox balance through its GEF-independent functions.
3. Relationship Between VAV1 and Diseases
3.1 Autoimmune and Chronic Inflammatory Diseases
Abnormal expression or dysfunction of VAV1 is associated with various autoimmune and chronic inflammatory diseases, such as rheumatoid arthritis, inflammatory bowel disease, and psoriasis. Therefore, VAV1 is considered a potential therapeutic target for these diseases.

In malignant tumors, such as breast cancer and lung cancer, overexpression of VAV1 is usually closely related to tumor malignancy, metastasis, and prognosis.

4. Rapid Detection of Small Molecules Mediating DDB1/CRBN Complex Interaction with VAV1 Based on TR-FRET
The interaction between DDB1/CRBN and VAV1 is detected using Eu-labeled anti-Tag1 antibody (TR-FRET donor) and Acceptor-labeled anti-Tag2 antibody (TR-FRET acceptor). Since the molecular glue MRT-6160 mediates the interaction between DDB1/CRBN and VAV1, the donor antibody and acceptor antibody are brought into proximity. Excitation of the donor antibody triggers fluorescence resonance energy transfer (FRET) to the acceptor antibody, causing the acceptor antibody to specifically emit a signal at a wavelength of 665 nm. This specific signal is proportional to the degree of interaction between MRT-6160 and DDB1/CRBN, VAV1. This homogeneous assay is simple to operate and requires no washing steps.
