1. Overview

AntBio MultiColor Flow Cytometry Mini Panel Kits are pre-optimized, ready-to-use reagents for multicolor flow immunophenotyping. Validated for human and mouse samples, they include 10+ classic immune panels for comprehensive immune cell identification, polarization profiling and functional subset quantification. All fluorophore combinations, antibody cocktails and staining protocols are fully verified. 2. Immunophenotyping Panels
2.1 Macrophage Panel
Macrophages are tissue-resident leukocytes differentiated from circulating monocytes, which originate from hematopoietic progenitor cells in the bone marrow. As professional phagocytes, both monocytes and macrophages participate in innate immunity (non-specific defense) and cell-mediated adaptive immunity (specific defense) in vertebrates. Existing as either sessile tissue-resident cells or migratory circulating cells, they execute phagocytosis to eliminate apoptotic cells, cellular debris and invading pathogens. Additionally, they activate lymphocytes and other immune cells to accelerate host immune responses against pathogens.
Note: This protocol outlines a universal detection strategy for total macrophages. Supplementary phenotypic markers are required to distinguish monocyte-derived infiltrating macrophages from tissue-resident macrophage subsets.

2.2 Natural Killer (NK) Cell Panel
NK cells are a major subset of innate lymphoid cells (ILCs) distinguished from adaptive T and B lymphocytes by lacking somatically rearranged antigen receptors encoded by recombination-activation genes. These large granular lymphocytes eliminate infected and tumor cells without pre-sensitization through coordinating activating and inhibitory receptors. Activated NK cells release perforin, granzymes and IFN-γ to kill target cells and modulate immune responses. Originating from bone marrow stem cells, they possess tissue-specific subsets and have promising applications in cancer immunotherapy.

2.3 T Cell Panel
T cells are thymus-matured adaptive immune lymphocytes with TCRs for MHC-restricted antigen recognition. CD4⁺ helper T cells regulate immune responses, while CD8⁺cytotoxic T cells kill abnormal cells. Memory T cells provide lasting protective immunity, and T cell-based therapies are widely studied for cancer treatment.

2.4 B Cell Panel
B cells are key adaptive immune cells that mature in bone marrow. They have unique rearranged B cell receptors (BCRs) to bind foreign antigens. When stimulated by antigens, B cells multiply and turn into plasma cells that make antibodies for humoral immunity; some become long-lived memory B cells for fast secondary protection. Aside from making antibodies, B cells present antigens to activate T cells and release cytokines to regulate immune reactions. Dysfunctional B cells cause autoimmune diseases, so they are important targets for immune disease therapies.

2.7 Dendritic Cell (DC) Panel
DCs are powerful antigen-presenting cells originating from bone marrow stem cells. They have unique tree-like extensions; immature DCs stay in skin and mucosal barriers to catch foreign antigens. After taking up antigens and maturing, DCs move to lymph nodes, present antigens with MHC molecules and activate naive T cells, connecting innate and adaptive immunity. DCs have two main subtypes: myeloid and plasmacytoid DCs with different roles. They trigger immune responses against germs and tumors while keeping immune balance. DCs are heavily studied to create new immunotherapy treatments.

2.8 Tissue-Resident Memory T (TRM) Cell Panel
TRM cells are stationary memory T cells, distinct from circulating Tcm and Tem subsets. They reside long-term in skin, lung, gut and mucosal tissues; markers CD69 and CD103 prevent their tissue exit. Re-encountered antigens prompt TRM cells to secrete IFN-γ, TNF and cytotoxic factors to locally clear pathogens and tumor cells rapidly. Local tissue niches sustain their long survival without persistent antigen exposure. TRM cells deliver frontline protection against infection and cancer, yet overactivation induces autoimmune inflammation. They are intensively studied for vaccine development and targeted cancer immunotherapy.

2.10 Mixed Lymphocyte Reaction (MLR) Panel
MLR is a classic lab immune test discovered in 1963. It mixes lymphocytes from genetically different donors to measure T cell responses to unmatched HLA proteins on antigen-presenting cells. There are two primary MLR types. In two-way MLR, both cell populations multiply. In one-way MLR, stimulator cells are treated with radiation or mitomycin C to stop their growth. After recognizing foreign MHC antigens, reactive T cells multiply and release cytokines such as IFN-γ. This process imitates the immune reaction that causes organ transplant rejection. Researchers commonly use MLR to judge transplant matching, find immune-regulating drugs, and check how well cancer immunotherapies work.
