Follow these steps to ensure smooth usage of the matrix gel.

一、Upon Receipt, Please Check

1.  Whether there is dry ice in the foam box;
2.  If the matrix gel is solid and the surface is smooth;
3.  If the color of the matrix gel is within the normal range (white/milky white without phenol red, yellow-pink-dark red with phenol red, determined by temperature changes);
4.  If you do not plan to use the matrix gel immediately upon receipt, store it in a -20/-80°C refrigerator for long-term preservation.

二、Operational Procedure Description

Before using the matrix gel each time, thaw it strictly according to the following procedure!

1、Pre-cool the "Matrix Gel Ice Box (abs7289)" (or prepare crushed ice) and a 200μL pipette tip box for more than 1 hour at -20°C. Before leaving work in the evening, take the matrix gel out of the -20/-80°C refrigerator and place it in the "Matrix Gel Ice Box" (or bury it in an ice box filled with crushed ice), then place the entire ice box in a 4°C refrigerator for overnight thawing;

2、The next day, after thawing is complete, you can use the matrix gel. Take the matrix gel out of the 4°C refrigerator and immediately place it in a pre-cooled -20°C "Matrix Gel Aliquot Ice Box" (ice box with crushed ice), then use it aseptically on a laminar flow hood/biosafety cabinet;

3、When using, take out the pre-cooled 200μL pipette tips from -20°C and open the matrix gel bottle cap (try to operate on the matrix gel thawing ice box);

4、After using the matrix gel, immediately put it back in the ice box and store it in a 4°C refrigerator for short-term preservation, use it up within two weeks.

 

三、Essential Notes

1、Do not directly contact the matrix gel with the palm of your hand;

2、The entire usage process should be carried out on the pre-cooled matrix gel aliquot ice box (ice box with crushed ice), and the remaining matrix gel can be stored at 4°C for up to two weeks;

3Never directly throw the matrix gel in the refrigerator for thawing and storage, always place it in the "Matrix Gel Ice Box" (or bury it in an ice box filled with crushed ice) before putting it in the 4°C refrigerator for thawing or storage; in addition, do not thaw or store it near the refrigerator door, or in a refrigerator that is often opened to take out reagents, try to thaw or store it inside/drawer of the 4°C refrigerator/air outlet;

4Since the matrix gel is sensitive to temperature, it is recommended to use a non-defrost type and stable temperature refrigerator for melting and storage, never thaw the matrix gel in a refrigerator with automatic defrosting;

5If using an ice box for overnight thawing, fix the position of the matrix gel bottle to ensure that the bottle will not tip over after the ice melts overnight, and the ice water will not submerge the matrix gel bottle mouth;

6Avoid repeated freeze-thaw cycles during the use of the matrix gel;

7Liquid matrix gel will quickly solidify above 8°C (especially high-concentration matrix gel). Therefore, when using, the consumables in contact with the matrix gel must be pre-cooled (such as pipettes, tips, EP tubes, etc.), and the entire experiment must be operated aseptically on ice;

8The matrix gel on ice should be in a liquid state when using, if the matrix gel has already solidified on ice, it indicates that the aliquot operation failed to maintain low temperature, causing the matrix gel to solidify, and should not be used again;

9If you find that the matrix gel in the 4°C refrigerator is very viscous but still liquid, you can place it in a pre-cooled -20°C ice box, put it in the -20°C refrigerator for 3-5 minutes, and the fluidity will generally return to normal; if it cannot be restored, it may be due to the refrigerator temperature being too high causing gelation, it is recommended not to use it.

Q1: Why is my matrix gel very viscous?
A1: The higher the protein concentration of the matrix gel, the more viscous the gel. If the concentration is higher than 13.0mg/mL, the matrix gel will be very thick. Matrix gel products are relatively viscous before dilution. The viscous high-concentration matrix gel can be used directly without dilution, such as for culturing tumor cells and/or angiogenic factors, injected into mice, cells can remain in place, facilitating in situ analysis and/or later excision; or diluted, used according to the standard concentration matrix gel product usage method, the specific dilution concentration is determined according to experimental needs.
A2: In addition to the product itself being highly concentrated and viscous, the state of the matrix gel is also related to temperature changes during transportation and storage conditions. Dry ice must be used throughout the transportation process. If the refrigerator where the matrix gel is stored has an automatic defrosting function, the temperature may rise during the defrosting process, causing the matrix gel to solidify. Therefore, never store matrix gel in such refrigerators with automatic defrosting. To ensure the effectiveness of the matrix gel, the number of freeze-thaw cycles should be minimized. Special attention: the temperature transfer of water, ice, ice water, and air is different, the air temperature transfer is particularly slow and the temperature distribution is uneven, therefore, the matrix gel thawing cannot be directly thrown in the 2-8°C refrigerator to thaw, it must be placed in an ice box/ice water mixture, then put in the 0-4°C refrigerator to thaw.
