Drug Conjugation
1. Sortase A-Mediated Enzymatic Conjugation (SMAC)
Sortase A recognizes the conserved LPXTG peptide motif and cleaves the bond between threonine and glycine residues. SMAC enables site-specific toxin attachment at predefined antibody loci to generate highly homogeneous, stable ADCs with uniform DAR values.

3. Complete Site-Directed Conjugation Kits
Step 1: Antibody Azide Modification
EndoS glycosidase trims antibody conserved N-glycans to expose terminal GlcNAc. Mutant β-galactosyltransferase (b.GalTY289L) transfers azide-functionalized GalNAz onto exposed GlcNAc via UDP-GalNAz substrate.

Copper-free SPAAC click chemistry conjugates cytotoxins, biotin or fluorescent probes to azide-modified antibodies.
