Evolved Antibody-drug Conjugates Raw Materials

Overview of ADCs

Antibody-Drug Conjugates (ADCs), known as "biological missiles", combine tumor-targeting specificity of monoclonal antibodies with potent cytotoxic payloads. ADC drug development encounters severe technical challenges across target identification, linker engineering, conjugation chemistry and preclinical pharmacodynamic characterization.

ANTBIO delivers a full-spectrum ADC research toolkit including raw materials, conjugation reagents and comprehensive detection kits to accelerate your ADC pipeline development.

Drug Conjugation

1. Sortase A-Mediated Enzymatic Conjugation (SMAC)

Sortase A recognizes the conserved LPXTG peptide motif and cleaves the bond between threonine and glycine residues. SMAC enables site-specific toxin attachment at predefined antibody loci to generate highly homogeneous, stable ADCs with uniform DAR values.

Table 2: Sortase A
Catalog No. Product Name
UA070086 Sortase A
  • High Catalytic Efficiency: Site-specific protein ligation with intact native structure and bioactivity
  • Sequence-Specific Recognition: LPXTG-dependent coupling compatible with glycine-modified acceptors and broad substrates
  • Mild Reaction Conditions: Physiological buffer conditions eliminate protein degradation risks
  • Tunable Reaction Kinetics: Adjust calcium concentration, substrate dosage and incubation time to optimize yield and conjugation efficiency

2. Glycan-Based Conjugation Toolkit

UA070042 Endo H (MBP Tag)
UA070039 Endo S
UA070055 Endo S2
UA070081 M-bovin-GalT1(Y289L)
  • Superior Homogeneity: Site-specific glycan modification achieves uniform drug loading for homogeneous ADC batches
  • Enhanced Stability: Preserves full antibody bioactivity; engineered glycans improve ADC structural stability and biocompatibility
  • Improved Therapeutic Window: Reduced off-target cytotoxicity and expanded safety margin for in vivo applications

3. Complete Site-Directed Conjugation Kits

Step 1: Antibody Azide Modification

EndoS glycosidase trims antibody conserved N-glycans to expose terminal GlcNAc. Mutant β-galactosyltransferase (b.GalTY289L) transfers azide-functionalized GalNAz onto exposed GlcNAc via UDP-GalNAz substrate.

Step 2: Cytotoxin Click Conjugation

Copper-free SPAAC click chemistry conjugates cytotoxins, biotin or fluorescent probes to azide-modified antibodies.
