Choosing the Right Biomaker Detection Platform for Drug Development: OneStep ELISA, StepSlim ELISpot and FCM Multiplex

Biomaker readouts sit at the center of modern drug development. IFN-γ release informs the efficacy and safety evaluation of CAR-T therapies; Th1/Th2 profiles track pharmacodynamic response in immunomodulation programs; and single-cell functional data reveal whether a candidate truly reprograms the immune response. Choosing the right detection technology — and the right kit format — directly affects data quality, throughput and sample economy.

OneStep ELISA: Four Formats Tuned to Different Screening Scenarios

The OneStep ELISA platform completes its assay within one hour through a simplified procedure — add sample and antibody cocktail, wash and add TMB substrate, add stop solution and read — while maintaining excellent sensitivity, a wide dynamic range and tight lot-to-lot consistency enabled by recombinant antibodies.

Within a single target such as human IFN-γ, the portfolio offers four purpose-built formats:

  • Standard format (S0C3005): high sensitivity, calibrated with international reference standards, suitable for residue detection.
  • OptiQuant Plus (S0C3100): IFN-γ is a core indicator for the release and efficacy evaluation of CAR-T therapies, reflecting activation and specific killing function; this kit is validated using samples from CAR-T clients.
  • Broad range (S0C3025): ultra-wide linearity eliminates the hassle of dilution, with a dynamic range of 0.80–1739.20 pg/mL (2,000-fold).
  • High sensitivity (S0C3055): ultra-high sensitivity down to 0.316 pg/mL for trace-level detection.

Where a workflow demands both trace sensitivity and an extended quantification window, the broad-range and high-sensitivity formats remove the two most common bottlenecks of routine ELISA: repeated sample dilution and signals buried below the detection limit.

StepSlim ELISpot: Functional Readout at the Single-Cell Level

ELISA reports the average biomaker concentration of a well; ELISpot reports how many cells are actually secreting. By capturing antigens released by individual cells as visible spots on a PVDF membrane, the StepSlim ELISpot series quantifies the frequency and function of biomaker-secreting cells — the readout that matters for vaccine studies, immune-monitoring and candidate immunomodulatory drug development.

The new-generation StepSlim kits were developed around four practical advantages:

  • Extreme sensitivity, authentic results: high-sensitivity paired antibodies enable efficient capture and detection, and the system is free of avidin — no endogenous biotin interference.
  • Ultra-fast workflow, doubled efficiency: enzyme-direct labeled antibodies save 1 hour of incubation, and ready-to-use pre-coated plates can be used straight out of the box, saving a full day of preparation.
  • Flexible customization: ready-to-use ALP substrate requires no on-site preparation, while the HRP series offers high catalytic efficiency; detachable and fixed plate formats are both available.
  • Stable and reliable outcomes: comprehensive validation covers data linearity, precision and competitor comparison analysis.

FCM Multiplex: Dozens of ELISA Tests in a Single Well

When the question is not one analyte but a biomaker network, the FCM multiplex bead assay measures multiple targets simultaneously from a single sample:

  • High-throughput parallel detection: multiple analytes measured in one well — one run equals dozens of traditional ELISA tests in efficiency.
  • Low-sample-volume friendly: only 25–50 μL of serum, plasma or cell supernatant is required, preserving precious samples.
  • High sensitivity and precision: limit of detection as low as 0.1 pg/mL; intra- and inter-assay CV < 5%, with data stability comparable to single-plex ELISA.
  • Flexible customization: free-combination target panels are supported.
