Background

ZNF512B (also known as KIAA1196 or GAM) is a transcription factor containing seven C2H2-type zinc finger domains and is highly conserved among vertebrates. Its core function involves specifically recognizing non-contiguous TTC motifs within satellite repeats in the genome via its zinc finger domains, directly recruiting SUV39H1/H2 methyltransferases to catalyze the formation of repressive histone H3K9me3 marks, thereby establishing and maintaining constitutive heterochromatin structure at pericentric regions. Additionally, ZNF512B can directly bind to the RBBP4 subunit of the NuRD chromatin remodeling complex through an internal FOG-1-like NuRD interaction motif (NIM), exerting transcriptional repression independent of its zinc finger domains. Recent studies have also revealed non-canonical functions of this protein in cell cycle regulation: it associates with the mitotic spindle to regulate mitotic exit, and its aberrant expression leads to severe metaphase arrest, subsequently affecting proper stem cell differentiation. With these complex and diverse functions, ZNF512B plays critical roles in transcriptional repression, higher-order chromatin structure formation, and cell cycle progression, and its dysregulation has been implicated in various pathological processes, including cancer and neurodegenerative diseases.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti- ZNF512B antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
