Background
ZMYND8 is a multi-domain transcriptional regulatory protein encoded by the ZMYND8 gene, playing critical roles in DNA damage response, transcriptional regulation, and chromatin remodeling. This protein contains a PHD zinc finger domain, a MYND domain, and a bromodomain, which enable it to recognize histone post-translational modifications (such as H3K4me0 and acetylated lysines) and sense cellular redox status. ZMYND8 recognizes distinct substrates through its different domains: its PHD zinc finger recognizes the "lysine 4 unmethylated" state on the histone H3 tail, the MYND domain binds to proteins such as the transcription factor BCL6, and the bromodomain binds to acetylated lysines. Functionally, ZMYND8 serves as a critical factor in DNA double-strand break repair—following DNA damage, it is phosphorylated by ATM/ATR kinases, recruiting the NuRD chromatin remodeling complex to damage sites to promote homologous recombination repair. Additionally, it exerts antioxidant protective effects by inhibiting the aryl hydrocarbon receptor (AhR) signaling pathway to reduce reactive oxygen species accumulation. Furthermore, ZMYND8 is closely associated with the development and progression of various cancers, including breast cancer, prostate cancer, and melanoma, influencing tumor proliferation and chemoresistance through regulation of oncogene expression or DNA repair pathways. Notably, ZMYND8 possesses both activating and repressive functions in regulating gene expression—for example, it can synergize through its PHD zinc finger and MYND domains to interact with histone deacetylase complexes and repress transcription of specific genes, while also promoting expression of other genes by recruiting histone acetyltransferases. This multifaceted nature makes ZMYND8 a potential target of great interest in the fields of tumor therapy, antioxidant defense, and tissue development.
