Background

YTHDF3 is a critical N6-methyladenosine (m6A) RNA modification "reader" protein that functions in the cytoplasm, where it specifically recognizes and binds to m6A-modified RNAs via its YTH domain, thereby regulating gene expression at the post-transcriptional level. Regarding its function, different perspectives exist within the scientific community: one view holds that YTHDF3 plays an auxiliary and synergistic role, cooperating with YTHDF1 to enhance the translation efficiency of m6A-modified mRNAs, or with YTHDF2 to promote their degradation, and it is also involved in the formation of stress granules; an alternative view, however, suggests that due to its highly similar structure and m6A-binding affinity to YTHDF1 and YTHDF2, the functions of these three proteins are redundant, collectively mediating the degradation of m6A-modified mRNAs. Despite ongoing debates over the precise mechanisms, the physiological and pathological significance of YTHDF3 is well established—it is upregulated in various cancers and promotes tumor progression, while also playing important roles in cardiac stress responses and antiviral immunity, making it a promising potential therapeutic target.
