Background
YAP is a highly conserved transcriptional coactivator encoded by the YAP1 gene and serves as the core downstream effector of the Hippo signaling pathway. This protein contains WW domains and a transcriptional activation domain, with its activity primarily regulated by phosphorylation modifications: when the Hippo pathway is activated, upstream kinases LATS1/2 phosphorylate YAP at multiple serine sites, promoting its cytoplasmic retention and degradation; when the pathway is off, dephosphorylated YAP translocates to the nucleus, where it binds to TEAD family transcription factors and initiates the expression of a series of target genes (such as CTGF, CYR61, BIRC5, etc.) that promote proliferation, inhibit apoptosis, and maintain stem cell characteristics. Physiologically, YAP is critical for organ size control and tissue regeneration; however, its aberrant hyperactivation is prevalent in various solid tumors (such as liver cancer, lung cancer, breast cancer, and colon cancer), driving tumor progression, metastasis, and drug resistance, making it a highly promising anticancer therapeutic target. In addition, YAP is also involved in mechanotransduction, sensing physical signals such as extracellular matrix stiffness and cell geometry, and plays important regulatory roles in tissue fibrosis and cardiovascular diseases.
