Background

WSB1 (WD repeat and SOCS box-containing protein 1) is an adaptor protein encoded by a gene on chromosome 17 in humans, functioning primarily as a substrate recognition subunit of the Cullin-RING E3 ubiquitin ligase complex (CRL). Its protein structure features two main functional modules: multiple WD40 repeats covering most of the sequence and a C-terminal SOCS box. The WD40 repeats are responsible for specifically recognizing and binding substrate proteins, while the SOCS box mediates interactions with core complex subunits such as Elongin B/C and Cullin-5, collectively catalyzing substrate ubiquitination and promoting their proteasomal degradation. Known degradation substrates of WSB1 include type 2 iodothyronine deiodinase (DIO2) and methylated RELA (p65), positioning it as a key player in regulating thyroid hormone metabolism and in terminating inflammatory responses in a timely manner through the degradation of NF-κB subunits. Furthermore, WSB1 exhibits broad tissue distribution, with higher expression levels in the spleen and lungs, and its dysfunction has been associated with various disease processes, including tumor proliferation and metastasis as well as neurological disorders.
