Background

WDR5 (WD repeat domain 5) is a nuclear protein of 334 amino acids with a molecular weight of approximately 36.6 kDa, belonging to the WD repeat protein family; its characteristic ring-shaped structure surface harbors two key binding sites, the WBM (WDR5-binding motif) and the WIN (WDR5-interacting) site, which mediate its interactions with nearly 200 proteins. The core function of WDR5 is to serve as a scaffold subunit of epigenetic regulatory complexes: it is a core component of the MLL/SET1 family histone methyltransferase complex and the NSL histone acetyltransferase complex, and by binding the N-terminus of histone H3 and recruiting partner proteins such as RBBP5 and ASH2L, it promotes H3K4 trimethylation, an epigenetic mark associated with transcriptional activation. Beyond its classical epigenetic functions, WDR5 also participates in regulating embryonic stem cell pluripotency, iPSC reprogramming, and chromatin accessibility at the Myf5 gene locus in skeletal muscle satellite cells. In clinical genetics, missense variants in WDR5 have been identified as the cause of a novel neurodevelopmental disorder, with patients exhibiting seizures, abnormal growth parameters, and skeletal and cardiac abnormalities; the variants cluster in specific surface regions of the protein, suggesting dominant-negative or gain-of-function mechanisms. In the oncology field, WDR5 is a highly regarded anticancer target, and small-molecule WDR5 inhibitors and degraders are under extensive preclinical evaluation; however, recent studies suggest that their anticancer mechanisms may be more related to ribosome-directed therapeutic effects than to the purely epigenetic regulation originally envisioned.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-WDR5 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti-WDR5 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
