Background

WDR5 (WD repeat-containing protein 5) is a highly conserved core subunit of the mixed-lineage leukemia (MLL/SET1) family of histone H3 lysine 4 (H3K4) methyltransferase complexes, playing a pivotal role in epigenetic regulation and gene transcription. Structurally characterized by its seven-bladed β-propeller domain formed by WD40 repeats, WDR5 serves as a critical scaffold that stabilizes the MLL complex and facilitates its interaction with specific target proteins, including the recognition of arginine-methylated histone tails and non-histone substrates. Beyond its canonical function in promoting H3K4 methylationa hallmark of active transcriptionWDR5 is implicated in diverse cellular processes such as cell cycle progression, embryonic development, and stem cell pluripotency maintenance. Its dysregulation and overexpression are frequently observed in various malignancies, particularly acute leukemias and solid tumors, where it acts as an oncogenic driver by sustaining the expression of key proliferative genes like MYC, thereby making WDR5 a promising therapeutic target for cancer treatment through the development of specific small-molecule inhibitors that disrupt its protein-protein interactions.
