Background
Vinculin is a highly conserved, ubiquitously expressed cytoskeletal adaptor protein that localizes to both cell-cell and cell-matrix adhesion sites, where it functions as a central regulator of mechanotransduction by coupling integrins to the actomyosin cytoskeleton through its head-domain binding to talin and its tail-domain binding to F-actin, PIP2 lipids, and multiple actin-organizing proteins, thereby stabilizing focal adhesions and adherens junctions in response to tension and phosphoinositide signals that trigger its transition from an auto-inhibited closed state to an active open conformation.
