IHC shows positive staining in paraffin-embedded human colon. Anti-VEGFA antibody was used at 1/100 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Background
Vascular Endothelial Growth Factor A (VEGF-A) is a 45 kDa homodimeric glycoprotein and the founding member of the VEGF family, encoded on chromosome 6p21.3 and expressed as at least seven splice isoforms (e.g., VEGF-A165) that are secreted or extracellular-matrix-bound to initiate angiogenesis and vascular permeability through binding VEGFR-2 on endothelial cells and neuropilin co-receptors, triggering PI3K-Akt, PLCγ-MAPK, and Src signaling cascades that drive endothelial proliferation, migration, and tube formation, while its transcription is up-regulated by hypoxia via HIF-1α and by growth factors such as PDGF and FGF, and its dysregulation underlies tumor neovascularization, diabetic retinopathy, and inflammatory disorders, making VEGF-A a pivotal therapeutic target for anti-angiogenic cancer drugs and anti-VEGF antibodies like bevacizumab.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human kidney.
Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti-VEGFA antibody was used at 1/2000 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
