Background

VDAC1 (Voltage-Dependent Anion Channel 1) is a key channel protein highly expressed on the outer mitochondrial membrane of eukaryotic cells and is a major member of the VDAC family. Structurally, it consists of 19 β-strands that form a transmembrane β-barrel structure, whose internal pore size can switch between "open" and "closed" states in response to changes in membrane potential, thereby regulating the transmembrane transport of various ions (such as Cl⁻ and K⁺) and metabolites (such as ATP, ADP, and pyruvate), serving as the central gateway for material exchange between mitochondria and the cytoplasm. Functionally, VDAC1 is far more than a passive channel; it dynamically interacts with a variety of ligands, including hexokinase and Bcl-2 family proteins, and is deeply involved in cellular energy metabolism, calcium homeostasis maintenance, and early regulation of apoptosis. When bound to pro-apoptotic proteins such as Bax, it can induce outer mitochondrial membrane permeabilization, releasing cytochrome c to initiate apoptosis, whereas anti-apoptotic proteins inhibit this process by occluding VDAC1. Consequently, aberrant expression or dysfunction of VDAC1 is closely associated with various diseases, including neurodegenerative disorders such as Alzheimer's disease and Parkinson's disease, myocardial ischemia-reperfusion injury, and various cancers, making it a popular target in drug therapy and disease diagnosis research.
