V5 Tag transfected 293T (Human embryonic kidney epithelial cell, Right panel) or 293T (Left panel) was stained with either Alexa Fluor® 647 Mouse IgG1, κ Isotype Control (Black line histogram) or SDT V5 Tag Mouse mAb (Alexa Fluor® 647 Conjugate) (Red line histogram) at 1/1000 dilution (0.1 μg), cells without incubation with primary antibody and secondary antibody (Blue line histogram) was used as unlabelled control. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
V5-tag is a short peptide tag for detection and purification of proteins. The V5-tag can be fused/cloned to a recombinant protein and detected in ELISA, flow cytometry, immunoprecipitation, immunofluorescence, and Western blotting with antibodies and Nanobodies.

Immunocytochemistry
ICC shows positive staining in Histone H3-V5 transfected 293T cells (top panel) and negative staining in vector-transfected 293T cells (below panel). Anti-V5 Tag (Alexa Fluor® 647 Conjugate) antibody was used at 1/200 dilution (meganta) and incubated overnight at 4°C. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).
