Background

USP5 (ubiquitin-specific protease 5) is a deubiquitinating enzyme belonging to the cysteine protease family. Its core structure is composed of a zinc finger ubiquitin-binding domain, a catalytic domain, and two tandem ubiquitin-associated domains, and it functions as a "recycler" of the ubiquitin pool. Unlike most other USP family members, USP5 specifically recognizes and cleaves unanchored polyubiquitin chains (i.e., free polyubiquitin not conjugated to substrate proteins), hydrolyzing isopeptide bonds to break down long chains into individual ubiquitin molecules, thereby maintaining the steady-state concentration of free ubiquitin monomers within the cell. During protein degradation, USP5 regulates proteasome function by preventing the accumulation of misfolded proteins and participates in DNA damage responses, cell cycle progression, and apoptosis through modulating the polyubiquitination status of key signaling proteins such as p53 and PTEN. Additionally, USP5 is involved in regulating the activity of autophagy receptor proteins (e.g., p62/SQSTM1), affecting autophagic flux efficiency. Aberrant expression of USP5 is closely associated with malignant progression and chemoresistance in various tumors (including glioma, colorectal cancer, and hepatocellular carcinoma), as well as with pathological processes in neurodegenerative diseases such as Parkinson's disease, making it a promising drug target for anticancer therapy.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-USP5 antibody was used at 1/250 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
